植物(wu)、土(tu)(tu)(tu)壤(rang)(rang)或(huo)(huo)(huo)(huo)者其他固(gu)體有機物(wu)樣(yang)(yang)品(pin)(pin)(pin)(如(ru)肉類等(deng))在(zai)(zai)進(jin)(jin)(jin)行(xing)(xing)有機碳(tan)δ13C和(he)有機氮δ15N同位(wei)(wei)素分析(xi)之(zhi)前須進(jin)(jin)(jin)行(xing)(xing)干燥(zao)、粉碎(sui)、酸化(hua)處理(堿性土(tu)(tu)(tu)壤(rang)(rang)的(de)情況下)等(deng)處理步驟(zou):采回的(de)樣(yang)(yang)品(pin)(pin)(pin)可(ke)以(yi)(yi)(yi)放在(zai)(zai)透氣性好、耐一(yi)定高溫(wen)的(de)器具(ju)或(huo)(huo)(huo)(huo)采樣(yang)(yang)袋中(zhong)(zhong),如(ru)玻璃或(huo)(huo)(huo)(huo)者陶瓷器皿(不能用(yong)(yong)報(bao)紙包(bao)裹烘(hong)(hong)干以(yi)(yi)(yi)免(mian)污染樣(yang)(yang)品(pin)(pin)(pin)),在(zai)(zai)干燥(zao)箱(xiang)中(zhong)(zhong)于(yu)60℃~70℃下干燥(zao)24 h~48 h(溫(wen)度不可(ke)太高,以(yi)(yi)(yi)免(mian)樣(yang)(yang)品(pin)(pin)(pin)碳(tan)化(hua)),研碎(sui),過(guo)60目(mu)或(huo)(huo)(huo)(huo)80目(mu)篩(shai),裝入密封小瓶、離心管或(huo)(huo)(huo)(huo)塑料袋中(zhong)(zhong)備用(yong)(yong),送檢樣(yang)(yang)品(pin)(pin)(pin)至少要求(qiu)0.2 g(最好在(zai)(zai)2 g以(yi)(yi)(yi)上,多些更(geng)好,以(yi)(yi)(yi)供備用(yong)(yong))。酸化(hua):測定堿性土(tu)(tu)(tu)壤(rang)(rang)中(zhong)(zhong)的(de)有機碳(tan)同位(wei)(wei)素,在(zai)(zai)干燥(zao)之(zhi)前需(xu)要進(jin)(jin)(jin)行(xing)(xing)酸處理,以(yi)(yi)(yi)消除(chu)土(tu)(tu)(tu)壤(rang)(rang)樣(yang)(yang)品(pin)(pin)(pin)中(zhong)(zhong)無(wu)機碳(tan)的(de)影響。具(ju)體步驟(zou)如(ru)下:(1)取適(shi)(shi)量(liang)研磨過(guo)篩(shai)后的(de)土(tu)(tu)(tu)壤(rang)(rang)樣(yang)(yang)品(pin)(pin)(pin)于(yu)小燒杯(bei)中(zhong)(zhong),加入適(shi)(shi)量(liang)濃度的(de)鹽酸(一(yi)般用(yong)(yong)0.5 mol L-1),由(you)于(yu)土(tu)(tu)(tu)壤(rang)(rang)中(zhong)(zhong)的(de)無(wu)機碳(tan)與(yu)鹽酸反應產(chan)生(sheng)CO2,所以(yi)(yi)(yi)有氣泡產(chan)生(sheng);(2)反應時間應不少于(yu)6 h,每隔1 h用(yong)(yong)玻璃棒攪拌一(yi)次(ci),使之(zhi)充分反應,以(yi)(yi)(yi)完全去(qu)除(chu)土(tu)(tu)(tu)壤(rang)(rang)中(zhong)(zhong)的(de)無(wu)機碳(tan),靜置(zhi),再倒掉(diao)上清(qing)液(ye);(3)用(yong)(yong)去(qu)離子水攪拌洗滌,靜置(zhi),倒掉(diao)上清(qing)液(ye),重(zhong)復3~4次(ci),以(yi)(yi)(yi)去(qu)除(chu)過(guo)量(liang)鹽酸,然后烘(hong)(hong)干備用(yong)(yong)。注意事項ATTENTION1.任何時候不能用(yong)(yong)裸露(lu)的(de)雙手觸(chu)摸樣(yang)(yang)品(pin)(pin)(pin)。若(ruo)用(yong)(yong)手操作,須帶無(wu)塵橡膠手套(tao)。2.烘(hong)(hong)干的(de)樣(yang)(yang)品(pin)(pin)(pin)要及時研磨或(huo)(huo)(huo)(huo)者保持干燥(zao),否(fou)則(ze)有返(fan)潮現象,給磨樣(yang)(yang)造(zao)成困難,而且會影響同位(wei)(wei)素比值。經(jing)過(guo)烘(hong)(hong)干的(de)樣(yang)(yang)品(pin)(pin)(pin)一(yi)般都需(xu)要粉碎(sui)才能進(jin)(jin)(jin)行(xing)(xing)分析(xi),為了(le)保證樣(yang)(yang)品(pin)(pin)(pin)的(de)均勻,粉碎(sui)...
