植物、土(tu)壤(rang)或(huo)者(zhe)其(qi)他(ta)固體有(you)機(ji)(ji)物樣(yang)(yang)品(pin)(pin)(pin)(pin)(如肉(rou)類等)在進(jin)(jin)行(xing)(xing)有(you)機(ji)(ji)碳(tan)δ13C和有(you)機(ji)(ji)氮δ15N同(tong)(tong)位(wei)(wei)素分(fen)析之前須進(jin)(jin)行(xing)(xing)干(gan)(gan)燥(zao)、粉(fen)碎、酸(suan)(suan)化處理(堿性(xing)土(tu)壤(rang)的(de)(de)(de)(de)情況下(xia))等處理步(bu)驟:采(cai)回的(de)(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)可以放(fang)在透氣(qi)(qi)性(xing)好(hao)、耐一(yi)(yi)(yi)定(ding)高溫的(de)(de)(de)(de)器具或(huo)采(cai)樣(yang)(yang)袋(dai)中(zhong)(zhong)(zhong),如玻璃(li)或(huo)者(zhe)陶(tao)瓷器皿(不(bu)(bu)能用(yong)(yong)(yong)(yong)報紙包裹(guo)烘干(gan)(gan)以免(mian)(mian)污(wu)染樣(yang)(yang)品(pin)(pin)(pin)(pin)),在干(gan)(gan)燥(zao)箱中(zhong)(zhong)(zhong)于60℃~70℃下(xia)干(gan)(gan)燥(zao)24 h~48 h(溫度(du)不(bu)(bu)可太高,以免(mian)(mian)樣(yang)(yang)品(pin)(pin)(pin)(pin)碳(tan)化),研碎,過60目或(huo)80目篩,裝入(ru)(ru)密(mi)封小(xiao)瓶、離心管(guan)或(huo)塑料袋(dai)中(zhong)(zhong)(zhong)備用(yong)(yong)(yong)(yong),送(song)檢樣(yang)(yang)品(pin)(pin)(pin)(pin)至少要(yao)求(qiu)0.2 g(最好(hao)在2 g以上,多些更(geng)好(hao),以供備用(yong)(yong)(yong)(yong))。酸(suan)(suan)化:測(ce)定(ding)堿性(xing)土(tu)壤(rang)中(zhong)(zhong)(zhong)的(de)(de)(de)(de)有(you)機(ji)(ji)碳(tan)同(tong)(tong)位(wei)(wei)素,在干(gan)(gan)燥(zao)之前需要(yao)進(jin)(jin)行(xing)(xing)酸(suan)(suan)處理,以消除土(tu)壤(rang)樣(yang)(yang)品(pin)(pin)(pin)(pin)中(zhong)(zhong)(zhong)無(wu)機(ji)(ji)碳(tan)的(de)(de)(de)(de)影(ying)響。具體步(bu)驟如下(xia):(1)取適量研磨過篩后的(de)(de)(de)(de)土(tu)壤(rang)樣(yang)(yang)品(pin)(pin)(pin)(pin)于小(xiao)燒杯中(zhong)(zhong)(zhong),加入(ru)(ru)適量濃度(du)的(de)(de)(de)(de)鹽(yan)(yan)酸(suan)(suan)(一(yi)(yi)(yi)般(ban)用(yong)(yong)(yong)(yong)0.5 mol L-1),由于土(tu)壤(rang)中(zhong)(zhong)(zhong)的(de)(de)(de)(de)無(wu)機(ji)(ji)碳(tan)與鹽(yan)(yan)酸(suan)(suan)反應產生CO2,所以有(you)氣(qi)(qi)泡產生;(2)反應時間應不(bu)(bu)少于6 h,每(mei)隔1 h用(yong)(yong)(yong)(yong)玻璃(li)棒攪拌(ban)一(yi)(yi)(yi)次,使之充分(fen)反應,以完全去除土(tu)壤(rang)中(zhong)(zhong)(zhong)的(de)(de)(de)(de)無(wu)機(ji)(ji)碳(tan),靜(jing)置,再倒掉上清液;(3)用(yong)(yong)(yong)(yong)去離子水(shui)攪拌(ban)洗滌(di),靜(jing)置,倒掉上清液,重復3~4次,以去除過量鹽(yan)(yan)酸(suan)(suan),然后烘干(gan)(gan)備用(yong)(yong)(yong)(yong)。注意事項(xiang)ATTENTION1.任何時候不(bu)(bu)能用(yong)(yong)(yong)(yong)裸露(lu)的(de)(de)(de)(de)雙(shuang)手(shou)觸摸樣(yang)(yang)品(pin)(pin)(pin)(pin)。若用(yong)(yong)(yong)(yong)手(shou)操作,須帶無(wu)塵(chen)橡膠手(shou)套(tao)。2.烘干(gan)(gan)的(de)(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)要(yao)及時研磨或(huo)者(zhe)保持(chi)干(gan)(gan)燥(zao),否(fou)則有(you)返潮(chao)現象,給磨樣(yang)(yang)造成(cheng)困難(nan),而(er)且會(hui)影(ying)響同(tong)(tong)位(wei)(wei)素比值。經過烘干(gan)(gan)的(de)(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)一(yi)(yi)(yi)般(ban)都需要(yao)粉(fen)碎才能進(jin)(jin)行(xing)(xing)分(fen)析,為了保證(zheng)樣(yang)(yang)品(pin)(pin)(pin)(pin)的(de)(de)(de)(de)均勻,粉(fen)碎...
