植物(wu)、土(tu)壤(rang)(rang)(rang)或(huo)者其他(ta)固體(ti)有(you)(you)(you)機(ji)(ji)物(wu)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)(如肉類(lei)等(deng))在進(jin)(jin)行(xing)有(you)(you)(you)機(ji)(ji)碳(tan)(tan)δ13C和有(you)(you)(you)機(ji)(ji)氮δ15N同位素分(fen)析(xi)之(zhi)前須(xu)(xu)進(jin)(jin)行(xing)干(gan)(gan)(gan)燥(zao)(zao)、粉碎(sui)(sui)、酸化處(chu)理(li)(li)(堿性(xing)土(tu)壤(rang)(rang)(rang)的(de)(de)(de)情況下(xia))等(deng)處(chu)理(li)(li)步驟(zou)(zou):采回的(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)可(ke)以(yi)(yi)放在透氣性(xing)好、耐(nai)一(yi)定(ding)高溫的(de)(de)(de)器具(ju)或(huo)采樣(yang)(yang)袋中(zhong)(zhong),如玻璃(li)或(huo)者陶瓷(ci)器皿(不(bu)能用(yong)(yong)報紙包裹烘干(gan)(gan)(gan)以(yi)(yi)免污染樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)),在干(gan)(gan)(gan)燥(zao)(zao)箱(xiang)中(zhong)(zhong)于(yu)60℃~70℃下(xia)干(gan)(gan)(gan)燥(zao)(zao)24 h~48 h(溫度不(bu)可(ke)太高,以(yi)(yi)免樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)碳(tan)(tan)化),研(yan)碎(sui)(sui),過60目或(huo)80目篩,裝入密封小(xiao)瓶、離心管或(huo)塑料袋中(zhong)(zhong)備用(yong)(yong),送檢(jian)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)至少要(yao)求0.2 g(最好在2 g以(yi)(yi)上,多些更好,以(yi)(yi)供(gong)備用(yong)(yong))。酸化:測定(ding)堿性(xing)土(tu)壤(rang)(rang)(rang)中(zhong)(zhong)的(de)(de)(de)有(you)(you)(you)機(ji)(ji)碳(tan)(tan)同位素,在干(gan)(gan)(gan)燥(zao)(zao)之(zhi)前需要(yao)進(jin)(jin)行(xing)酸處(chu)理(li)(li),以(yi)(yi)消除(chu)土(tu)壤(rang)(rang)(rang)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)中(zhong)(zhong)無(wu)機(ji)(ji)碳(tan)(tan)的(de)(de)(de)影(ying)響。具(ju)體(ti)步驟(zou)(zou)如下(xia):(1)取適量(liang)(liang)研(yan)磨過篩后的(de)(de)(de)土(tu)壤(rang)(rang)(rang)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)于(yu)小(xiao)燒杯中(zhong)(zhong),加(jia)入適量(liang)(liang)濃度的(de)(de)(de)鹽(yan)(yan)酸(一(yi)般用(yong)(yong)0.5 mol L-1),由于(yu)土(tu)壤(rang)(rang)(rang)中(zhong)(zhong)的(de)(de)(de)無(wu)機(ji)(ji)碳(tan)(tan)與(yu)鹽(yan)(yan)酸反(fan)(fan)應(ying)產生CO2,所以(yi)(yi)有(you)(you)(you)氣泡(pao)產生;(2)反(fan)(fan)應(ying)時間應(ying)不(bu)少于(yu)6 h,每隔1 h用(yong)(yong)玻璃(li)棒(bang)攪拌一(yi)次,使之(zhi)充分(fen)反(fan)(fan)應(ying),以(yi)(yi)完全去(qu)除(chu)土(tu)壤(rang)(rang)(rang)中(zhong)(zhong)的(de)(de)(de)無(wu)機(ji)(ji)碳(tan)(tan),靜置,再(zai)倒(dao)(dao)掉(diao)上清液;(3)用(yong)(yong)去(qu)離子水攪拌洗滌,靜置,倒(dao)(dao)掉(diao)上清液,重復3~4次,以(yi)(yi)去(qu)除(chu)過量(liang)(liang)鹽(yan)(yan)酸,然后烘干(gan)(gan)(gan)備用(yong)(yong)。注意事項ATTENTION1.任(ren)何時候(hou)不(bu)能用(yong)(yong)裸露的(de)(de)(de)雙(shuang)手(shou)觸摸樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)。若(ruo)用(yong)(yong)手(shou)操作,須(xu)(xu)帶無(wu)塵(chen)橡(xiang)膠(jiao)手(shou)套(tao)。2.烘干(gan)(gan)(gan)的(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)要(yao)及(ji)時研(yan)磨或(huo)者保持干(gan)(gan)(gan)燥(zao)(zao),否則有(you)(you)(you)返潮現象,給磨樣(yang)(yang)造成困難,而且會影(ying)響同位素比值。經過烘干(gan)(gan)(gan)的(de)(de)(de)樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)一(yi)般都需要(yao)粉碎(sui)(sui)才能進(jin)(jin)行(xing)分(fen)析(xi),為(wei)了保證樣(yang)(yang)品(pin)(pin)(pin)(pin)(pin)的(de)(de)(de)均勻(yun),粉碎(sui)(sui)...
